Total glucans within a sample are solubilized in ice-cold 12 M H2SO4 and hydrolyzed to near completion in 2 M H2SO4. Remaining glucan fragments are quantitatively hydrolyzed to glucose using a mixture of exo-1,3-βglucanase and β-glucosidase. An additional sample portion undergoes hydrolysis of the α-Glucans to glucose using amyloglucosidase plus a-amylase. The glucose in both sample test portions are measured using GOPOD reagent. β-Glucan is determined by difference between the total glucans and α-glucans.

Unfortunately, this test only reports Beta-glucan.
Tinture samples must have enough beta-glucan present to be dected by this method. Please view current LOQ/LODs on the assay's tech data sheet.
