Technical data sheet
- Method description
This method determines the amount of Vitamin D3 (cholecalciferol) in various food products. An internal standard is added, and the sample is saponified in ethanolic KOH. It is then extracted into a heptane organic phase and evaporated to dryness. The sample extract is reconstituted and fractions of the internal standard and D3 are collected using a solid phase extraction (SPE) column. The fractions are then concentrated, and the level of D3 is determined using LC-MS/MS.

Our standard turnaround time is 10 business days for most assays. There are some assays that require a longer turnaround time. We also offer a RUSH service that is half the time of the standard turnaround time of the assay at double the cost of the assay. A few assays that we provide cannot be rushed due to the nature of the test. Please check the specific assay you are interested in regarding the ability to RUSH the turnaround time.

Vitamin D can be susceptible to heat exposure. It is best practice to ship samples on the cool side during warmer months, and ensure that the sample is not subjected to extreme temperatures or temperature swings.
If the product is fortified, the ingredient list will provide important information on the Vitamin D source. Ergocalciferol is the most common Vitamin D2 fortified ingredient, and Cholecalciferol is the most common Vitamin D3.
If the product is fortified and/or contains naturally occurring Vitamin D, then it can be helpful to examine the potential sources of Vitamin D. Vitamin D3 is naturally found in animal products such as fish, eggs, and meat. Very few foods have naturally occurring Vitamin D2, however, some types of mushrooms may have Vitamin D2.
